MicroRNA Profile in Treatment-Naïve Adults with Primary Progressive Multiple Sclerosis
In a study of 20 treatment-naïve patients with primary progressive multiple sclerosis (PPMS) and 25 healthy controls, 26 microRNAs showed significant differences between the groups. After correction for false discovery rates, 24 of these differences remained significant.

Why it matters
Identifying specific microRNAs associated with PPMS could enhance our understanding of the disease's biological mechanisms and aid in the development of potential biomarkers, but further research is necessary to confirm these findings.
What this does not prove
Larger, longitudinal studies are required before diagnostic or mechanistic conclusions can be drawn.

study phaseMETHODS AND RESULTS: In this exploratory case-control study, plasma samples from 20 newly diagnosed, treatment-naïve patients with PPMS and 25 healthy controls with broadly comparable age and sex distributions were assayed for 84 neurologically relevant miRNAs using high-throughput real-time quantitative polymerase chain reaction on the BioMark HD platform.

relevant ms typesBACKGROUND: Primary progressive multiple sclerosis (PPMS) is a disabling phenotype marked by gradual neurological worsening from disease onset and limited biomarker availability.

findingsTwenty-six miRNAs exhibited nominal between-group differences, of which 24 remained significant after Benjamini-Hochberg false discovery rate correction at q < 0.05.

mechanismsIntegrated analyses associated the biologically prioritized target set with miRNA biogenesis, stress-responsive signaling, inflammatory pathways, hypoxia/angiogenesis, and cell-cycle regulation.
