Immune Response Evaluation in vaccinated Persons with MS across DMT Classes
In a study involving 183 vaccinated individuals with multiple sclerosis (PwMS), distinct patterns of immune response to SARS-CoV-2 were observed based on the class of disease-modifying therapy (DMT) used. Notably, B-cell depleters showed lower Spike IgG responses, while S1PR modulators consistently exhibited lower antibody and T-cell responses. Those on fumarate treatments had diminished cellular responses in later assessments.

Why it matters
These findings suggest that different DMTs may significantly influence immune responses to vaccinations, emphasizing the need for personalized approaches to vaccination strategies in PwMS.
What this does not prove
The study does not establish causality between DMTs and immune responses due to its observational design.

primary endpointOBJECTIVE: Evaluate longitudinal SARS-CoV-2 Spike IgG and antigen-specific T-cell responses in vaccinated PwMS across DMT classes in a decentralized study.
findingsCONCLUSION: DMT class was associated with distinct humoral and cellular SARS-CoV-2 immune-response profiles in vaccinated PwMS, supporting multidimensional immune assessment and decentralized longitudinal biospecimen collection in MS research.
